Identification of Celery mosaic potyvirus, study its biological properties and identified its vector

Abstract

Celery mosaic potyvirus (CeMV, Genus: Potyvirus, Family: Potyviridae) was identified on the basis of symptoms on indicator plants, Apium graveolens L., Datura stramonium L., Datura metel L., Hibiscus esculentum L., Vigna unguiculata L., Chenopodium amaranticolor Cost & Reyn., mechanically inoculated by extract from Apium graveolens L. showing mottle and yellow streak symptoms. Vein clearing turned to yellow mottle then to yellow streaks on the leaves associated with color breaking-like pattern, leaves deformations and stunting of entire plant were manifested on inoculated A. graveolens plants. D. stramonium was found exhibited mosaic symptoms on uninoculated leaves 8 days after inoculation, then small chlorotic spots developed on the other leaves accompanying with green vein banding. D. metel responded to the inoculation by mosaic after 8 days of inoculation. Small chlorotic spots were developed on the inoculated leaves of Ch. amaranticolor after 10 days of inoculation, chlorotic spots on the uninoculated leaves of Hibiscus esculentum after 14 days of inoculation and systemic mosaic on Vigna unguiculata after 10 days of inoculation were observed. The dilution end point, thermal inactivation point, longevity invitro for the virus were found to be, 10-2 – 10-3, 60˚C, and 6 days respectively. Mottle and yellow streak symptoms were manifested on A. graveolens leaves exposed to viruliferous aphids Myzus persicae Sulz., Aphis faba, Aphis gossypii for 1 minute (aphids were given an acquisition period of 15 minutes). CeMV was found to be transmitted by A. graveolens and D. metel seeds.
