Effect or purified lawsone and aqueous extract of henna (lawsonia inermis) leaves on some pathogenichacteria and their Beta-lactamase activity

Abstract

The Lawsone has been extracted from henna (lawsonia inermis) leaves, the extract was purified by using two phase system and the components of the extract were identified by thin layer chromatography. Melting point of lawsone and the absorption characteristics in the ranges of infra red and ultraviolet spectrum were also identified. The extracted lawsone antibacterial effects against some pathogenic types of gram positive and gram negative bacteria including Psedomonas aerugionsa, Escherichiade coli, and Staphylococcus aurous has been studied, and the results showed that Lawsone has high inhibitory effect against β-lactamases extracted from investigated isolate. The comparison between the inhibitory effect of purified lawsone and aqueous extract was also made. The comparison showed that, the inhibitory effect of the 50 mg of purified lawsone against Psedomonas aerugionsa, Escherichia coli, and Staphylococcus aurous were identified in surrounded zones with the diameter of 18, 17, and 25 mm respectively, while the diameters of zones with aqueous extract were 20, 10, and 11 mm.Plasmid characteristics of different isolates that have been studied using salting out method followed by gel agarose electrophoresis revealed that each isolate type contained one large and 2-5 small plasmids. Different sources of plasmid showed relatively different plasmids sizes.
