Abstract

The aim of this study was to isolate the Staphylococcus species as essential causative agents for the chronic otitis media . 150 swab were collected from patient with chronic otitis media and cultured in different media. (62) species of Staphylococcus were isolated , it is found that Staphylococcus negative coagulase - positive mannitol is dominat (n=33) 53.22% . Staph. xylosus gave the higher rate of isolation with higher percentage (32.25 %) , the latter shows higher ability to produce thicker biofilm with (50%) . These isolates were subjected to biochemical test and Api-staph ( bioMerieux) system and observation their ability to produce biofilm invitro. Mannitol salt agar with different concentration of NaCl (10%,15% and 20% ) was used to determine the ability of different species of Staph. to tolerance high salt concentration, Antimicrobial susceptibility was achieved by disc diffusion method . Well method used to determine the effect of some topical antiseptic . All isolations show high ability of growth at highly concentrated NaCl media . The results obtained have shown that the 15% and 20% of NaCl concentration are the most dominant in the inhibition of the biofilm production . The outcome of this research work show that the Ciprofloxacin is the most effective antimicrobial against all isolates with percentage 93.54% . On the other hand the Gention violet solution (0.5%) is found to be the most effective topical antiseptic in preventing the growth of bacteria with high means of inhibition zone (28.1 16.5 )mm Staph. capitis and with high ability to prevent bacteria to biofilm formation
