The Effect of Continuous Darkness and Continuous Light on the Reactivity of Smooth Muscles to Drugs in the Rat Vas Deferens

Abstract

Background: The vasoconstricting agents: nor-adrenaline and 5- hydroxytryptamine (5-HT) have a stimulant action on smooth muscle contractility of the rat vas deferens.Objective: This study aimed to investigate the effect of exposure to continuous darkness and continuous light on the contractility of the vasa deferntia smooth muscles from rats to applied nor-adrenaline and 5-HT.Method: Male albino wistar rats were divided into 3 experimental groups. Group 1: Control animals, were exposed to the ordinary photoperiod each day. Group 2: Rats were kept in a dark room. Group 3: In a room under a bright artificial light.All animals were killed after 4 weeks.Results: Vasa deferentia preparations from continuous dark group of rats exhibited a reduced reactivity with a significant lower maximal response to 5-HT than those from control rats. The maximal response in the control vasa deferentia preparations were nearly doubled compared with that of the continuous dark preparation where as they responded to exogenous nor-adrenaline with no significant difference from those of continuous dark animals. Vasa deferentia of continuous light animals responded with decreased reactivity and a smaller maximal response to both vasoconstrictors compared with that of control animals.Conclusion: It is concluded that changes in the rhythm of the photoperiod have considerable effects on the reactivity of the vasa deferentia smooth muscle from rats to applied nor-adrenaline and 5-HT.

Abstract

¬ Background: Adenosine mediates homodynamic changes and resulted in the production of acute renal failure (ARF) in female Albino-Wister rats, therefore, adenosine level increases highly in ARF. Objective: This experiment was designed to investigate the effect of the adenosine antagonist aminophylline and the adenosine agonist indom-ethacin on glycerol-induced ARF.Method: Glycerol induced ARF was produced by a single dose (10ml/kg, 50%v/v with distilled water i.m) in rats, which were restricted to drinking water. Aminophylline was used in our study in a dose of 25mg/kg, i.p) while the dose of indomethacin was 10mg/kg, i.p), assessment of renal function was done by measuring blood urea nitrogen (BUN), serum creatinine (Scr), and creatinine clearance (Ccr)Results: Aminophylline exert its protective effect against glycerol induced-ARF by lowering the elevation in concentrations of BUN, Scr, and mortality rate with a markedly attenuation of the sever impairment Ccr. On the other hand, indomethacin potentiates glycerol induced-ARF by significantly increase the elevation in concentrations of BUN, Scr and mortality rate with severely suppressed the decrement in Ccr.Conclusion: Aminophylline has ameliorating effect on glycerol induced ARF while the prostaglandin inhibitor indomethacin has reversed effect.

Abstract

Melatonin is the major secretory product of the pineal gland, playing anintegral role in numerous metabolic, physiological and behavioral processes.This study represents an attempt to understand the effect of environmentalchanges on the responses of a selected isolated tissue to pharmacologicallyactive substances in certain species which may lead to important experimentaland clinical implications in future. Twenty male albino wistar rats were dividedinto three experimental groups:Group 1: Control animals, comprising 6 rats were exposed to 12:12 hour light:dark cycle for 4 weeks.Group 2: Composed of 8 rats, was kept in a dark environment for 4 weeks.Group 3: Consisted of 6 rats aged 6-7 weeks, subjected to continuous darknessfor 2 weeks, at the end of the first week the rats treated daily with s.c melatonininjection at 1.00 p.m. for 7 consecutive days. The result of this study showedthat the vas deferens preparation from the third group reacted with significantincrease in reactivity to nor-adrenaline and 5-HT than that of control andcontinuous dark animals.The limitation of time and shortage of materials may be blamed for thekind and quantity of results obtained and the conclusions made. Therefore,further studies with regard to the exposure period have to be undertaken infuture to extensively elaborate on this point.Key words: Melatonin, Vas deferens, 5-HT, Nor-adrenaline.

الخلاصةالظلام المطوَل له تأثیرات فسلجیة وكیمیائیة ومرضیة على أعضاء وأجهزة ووظائف الجسمالمتنوعة. أجریت هذه الدراسة على الحیوانات المختبریة (الجرذان) بقصد التحري عن التأثیرات المحتملةللتغیرات البیئیة على تفاعلیات العضلة الملساء للقنوات الدافقة في المجامیع المتشابهة الأعمار بالإضافةإلى استعمال مركب المیلاتونین ودراسة تأثیره علیها. تم تقسیم الجرذان إلى ثلاث مجامیع: المجموعةالأولى تم تعریضها إلى ۱۲ ساعة من الظلام و ۱۲ ساعة من الضوء لمدة أربعة أسابیع. المجموعةالثانیة حفظت في بیئة مظلمة ولمدة ٤ أسابیع ایض اً. المجموعة الثالثة تم تعریضها إلى ظلام مستمرولمدة أسبوعین مع حقنها بمركب المیلاتونین خلال الأسبوع الثاني فقط.وجد ان القنوات الدافقة المأخوذة من المجموعة الثالثة أحدثت زیادة واضحة في تفاعلاتها لعقارالنورأدرینالین و- 5 هیدروكسیتربتامین بالمقارنة مع الأنسجة المأخوذة من حیوانات السیطرة والحیواناتالمعرضة للظلام المستمر لمدة ٤ أسابیع, قد تعزى هذه الزیادة إلى التأثیرات المباشرة او غیر المباشرةلمركب المیلاتونین من خلال إفراز النورأدرینالین من النهایات العصبیة السمبثاویة او قد تحدث بسببتركیب عدد جدید من المستقبلات
