Abstract

Summary:
Background: Mitochondria play an important role in the regulation of physiological type of cell death (Apoptosis) this type of cell death can be stimulated by two major pathways: external (Fas-Fas ligand interaction) and internal mitochondrial pathway which require disruption of mitochondrial transmembrane potential (∆طm ) which leads to opening of mitochondrial channels that lead to release of cytochrome –C which would execute apoptotic process.
Patients and Methods:The study included 32 normal subjects; 2 ml of venous blood were aspirated from each of them and processed for peripheral blood lymphocytes separation (PBL)
Then lymphocyte apoptosis was studied before and after exposing (PBL) to hyperthermia by these three methods to compare which of them can detect earliest apoptotic changes: Cell count and viability, morphological changes (by acredine orange stain) and the changes in mitochondrial membrane potential by Mitolight Apoptosis Detection Kit.
Results: Our results showed that the percentage of lymphocyte apoptosis detected by mitolight was significantly higher than those detected by other two methods.
Conclusion: This study shows that mitochondrial changes can detect earlier apoptotic process than morphological features detected by DNA stains.

Abstract

ABSTRACT:BACKGROUND:The development and progression of diabetic nephropathy is dependent on glucose homeostasis and many other contributing factors. Diabetic nephropathy is a leading cause of end-stage renal failure, accounting for 35 to 40% of all new cases that require dialysis therapy worldwide. Recent clinical studies clearly demonstrated that hyperglycemia and oxidative stress is an important causal factor in mediating the development and progression of diabetic kidney disease.OBJECTIVE:To evaluate the therapeutic effect of levamesole in diabetic nephropathy .METHODS:The study included 10 rabbits Weight (1kg ±20 gm); they were followed up for 7months. Blood was aspirated from marginal ear vein after agitation with xylol for estimation of fasting blood glucose and malondialdehyde (MDA) which is used as marker of oxidative stress. Rabbits were given 110 mg / kg alloxan to induce diabetes. In the second month rabbits became diabetic without development of nephropathy. After 1 month from being diabetic a bosture dose of alloxan was given (125 mgkg). After 1 month of the bosture dose blood glucose level further increased and rabbits developed albumin urea. Once rabbits developed albumin urea they received levamesole 2 mgkg EOD for 6 weeks.RESULTS:The study results showed that diabetic nephropathy is associated with high blood glucose level (300-400mgdl) and oxidative stress (significant increase in MDA level). The nephropathy (albumin urea) and oxidative stress can be reversed by levamesole.CONCLUSION:The antioxidant effect and immune modulating properties of levamesole provided a protective therapy against the development of diabetic nephropathy.KEY WORDS: diabetic nephropathy, levamesole, MDA.

Abstract

Background: Asthma is an inflammatory airway disease; this inflammatory response can
be attributed to reduced lymphocyte apoptosis in peripheral blood and in airway tissues.
The mechanism behind this could be attributed to decreased Bcl2 protein and increase
Bax protein in peripheral blood lymphocytes of asthmatic patients.
Aim: to explore the mechanism behind decreased lymphocyte apoptosis in peripheral
blood of asthmatic patients at cellular level.
Method: Ninety four subjects; (44) control and (50) patients were included in this study
during the period from (2003) to (2004). The aspirated lymphocytes for each individual
were prepared and stained by immunocytochemistry to study the percentage of anti-
apoptotic Bcl-2 protein, pro-apoptotic Bax protein and their ratio.
Results: Our results showed that the percentage of Bcl2 protein in peripheral blood
lymphocyte of asthmatic patients were significantly higher than control (33.23±11.56,
21.47±4.16)(P=<0.00001).
The percentage of Bax protein in peripheral blood lymphocyte in asthmatic patients was
significantly lower than the control (21.43±6.88, 24.89±4.8) (P-O.0323). Bcl2/Bax ratio
was positive in asthmatic patients there was significant difference in comparison with the
control (1.68±0.57, 0.87 ± 0.17) (P=<0.00001).
Conclusion: This study clarifies the role of these 2 proteins in the process of reduction of
apoptosis in peripheral blood lymphocytes of asthmatic patients.

The effect of blood lead level on packed cell volume in a sample of Iraqi population.

Abstract

Background: Low hemoglobin levels and lead exposure remain significant health issues in many parts of the world, often occurring together, however most of the researches concentrate on the effect of high levels of lead .This study aimed to demonstrate the effect of blood lead level on hemoglobin and packed cell volume (PCV).Patients and Methods: Fifty eight adult volunteers were enrolled in this study with age range between 23 and 55 years, working in the different departments in medical college of Baghdad University. Whole blood was withdrawn in EDTA tubes. Hemoglobin and PCV were measured and blood lead level was estimated using blood lead testing system including lead care kit and lead care analyzer.Results: After measuring blood lead level and correlate it to PCV in all volunteers results revealed a significant negative correlation between them which is obvious in both smokers and non-smokers groups. Age was found to correlate positively and significantly with blood lead level in non smokers group. Comparing the blood levels of lead and PCV in both smokers and non-smokers shows non-significant difference between them although blood lead level was higher in smokers group.Conclusion: Blood lead levels is increased with age. There is a significant decrease in PCV with the increase in blood lead level which is noticed in both smokers and non smokers. There is a significant increase of blood lead levels in smokers group.Key words: blood lead, packed cell volume, age, smoking.

الخلاصة:خلفية الدراسة: تبقى قلة مستوى خضاب الدم والتعرض للرصاص يبقى مشكلة صحية في اجزاء من العالم .مع ذلك فان اكثر الابحاث تركز على تاثير المستوى العالي للرصاص في الدم .الاهداف: ان الهدف من هذه الدراسه بيان تاثير مستوى الرصاص القليل على خضاب الدم و نسبة خلايا الدم الحمراء.الطرائق والمرضى: شملت الدراسة ثماني وخمسين مريضا متطوعين تتراوح اعمارهم بين 23-55 سنة .و تم قياس نسبة خضاب الدم ونسبة خلايا الدم الحمراء والرصاص في الدم بواسطة جهاز خاص لذلك. وتم سحب الدم في انابيب اي دي تي اي(EDTA).النتائج: اظهرت النتائج وجود علاقة معنوية بين نسبة الرصاص في الدم ونسبة خلايا الدم الحمراء في كلا مجموعتي المدخنين وغير المدخنين وكذلك هناك علاقة معنوية بين العمر ومستوى الرصاص في الدم.الاستنتاج: نستنتج من هذه الدراسة وجود علاقة ذات دلاله احصائية بين مستوى الرصاص في الدم والعمر ونسبة الخلايا الحمراء في المدخنين وغير المدخنين.كما ان نسبة الرصاص في دم المدخنين عالية بدلاله احصائية. مفتاح الكلمات: الرصاص في الدم ,حجم كريات الدم الحمر , العمر , التدخين
