Evaluation of the antibacterial activity of human cathelicidin peptide-LL-37 in the presence of acidified nitrite

Abstract

Background and objective: Bacterial resistance to conventional antibacterial agents hasincreased recently and this resistance results in complicated infections. Multiple protectivemechanisms can evolve in mammalians to maintain the body protected from infections.Human cathelicidin antimicrobial peptide LL-37 and acidified nitrite are importantcomponents of the innate immune system that partake in preventing infections. Cathelicidinpeptide LL-37 can be produced by epithelial tissues as well as by macrophages aftermicrobial infections. This study was carried out to evaluate the antibacterial activity ofLL-37 and acidified nitrite (AN) both individually and combined for their effect against thestandard the strains of E.coli ATCC 25922 and S. aureus ATCC 25923.Methods: Flat-bottom micro well plates (96 wells) were used for the determination ofbacteriostatic activity. Sodium nitrite (NaNO2) and ascorbic acid (AA) were used to produceacidified nitrite (AN). The singular and combined forms of the antibacterial agents wereused for evaluating the antibacterial activities of LL-37 through estimation optical densityvalues at 480nm (OD480nm).Results: The LL-37 peptide showed antibacterial activity against E.coli ATCC 25922 andS. aureus ATCC 25923. The antibacterial efficacy was enhanced when the peptide wastested in combination with AN (P <0.001). In contrast, the combination of LL-37 withNaNO2 and AA has an antagonistic effect (P <0.001) on its antimicrobial properties.Conclusion: The combination of LL-37 with AN has a synergistic on the peptide’santimicrobial effect. Therefore, LL-37 which might show little antibacterial activitywhen used alone can provide protection when used in combination therapy with otherantimicrobial agents.

Extended-spectrum beta-lactamases (ESBLs) detection in some uropathogenic bacteria and their correlation with biofilm formation

Abstract

Background and objective: The tolerance of biofilms to antibiotics results in the dissemination of resistance. Many of the recurrent urinary tract infections are assumed to be caused by biofilm producing uropathogenic isolates. This study aimed to investigate the correlation between the ability of biofilm formation and extended spectrum beta lactamase producing uropathogens.Methods: A total of 438 urine specimens were collected from Rizgary Teaching hospital in Erbil, Iraq, from September to December 2017. Extended spectrum beta lactamase was determined using the Vitek-2 automated system and confirmed by using the combination disk diffusion test. Biofilm formation was checked using 96-well flat bottom microtiter plates.Results: Out of the 438 urine specimens, only 37.89% (n =166) developed an infection, the most common isolate was Escherichia coli. The distribution of the bacterial species according to the patients' gender found to be significant (P = 0.014). The proportion of betalactamase producing isolates was 29% (n = 31). The strength of biofilm formation among Klebsiella species was significantly higher than in Escherichia coli (P <0.001), while a non-significant difference (P = 0.163) was observed between beta lactamase production and ability of biofilm formation. The sensitivity and specificity of VITEK-2 in the detection of extended-spectrum beta-lactamase were 79.48% and 80.95%, respectively.Conclusion: The study revealed that Klebsiellae species were stronger biofilm producers. Beta lactamase producing isolates do not have a greater ability of biofilm formation.
