Preparation and potentiometric study of phenytoin selective electrodes based on a PVC matrix membrane

Abstract

Phenytion selective electrodes were prepared based on a complex phenytion-phosphomolybdate as an active material using the plasticizers di-octyl phthalate (DOPH), tri-butyl phosphate (TBP), o-nitro phenyl octyl ether (ONPOE) and di-butyl phthalate (DBPH) in a PVC matrix membrane. The properties of the prepared electrodes were studied, such as: slope, concentration range, detection limit, lifetime, pH effect and selectivity. The experimental results showed that the best electrode was based on DOPH and DBPH as plasticizers, displaying a linear range from1.00 x 10−4 M to 1.00 x 10−1 M and 1.00 x10−1 M to 5 x10−4 with a Nernstian slope of 58.5 mV/decade and 55.8 mV/decade, correlation coefficient of 0.9997 and 0.9998, The detection limit was 5.5 x 10−5 M and 8.0 x10−5, the lifetime was around 60 and 45 days respectively. The proposed electrodes were successfully applied to the determination of phenytion in a pharmaceutical preparation.

تم تحضير عدة أقطاب ايونيه في مادة PVC حساسه للفينتون بالاعتماد على المعقد (Phenytoin-phosphomolybdic acid) كماده فعاله. هذه الماده الفعاله تكون مذابه في عدة مواد ملدنه منها , Di-butyl phthalate (DBPH), Tri-butyl phosphate (TBP), O-nitrophenyloctylether (ONPOE), Di-octylphthalate (DOPH),تم مزج احد هذه الملدنات والمعقد مع PVC لتكوين الغشاء ومن خلال منحنى التدرج تم دراسة خواص هذه الاقطاب والتي تشمل (منحنى الانحدار و مدى التركيز وحد التحسس و عمر القطب و تأثير ال pH ) ومن خلال الدراسه وجد ان الأقطاب المتكونه من (DBPH,DOPH) كمواد ملدنه تمتلك مواصفات جيده يمكن الأعتماد عليها في تعيين الفينتون بصوره دقيقه, منحنى الأنحدار لهذه الأقطاب كان يساوي تقريباً (55.8,58.5 (mV/decade مدى التركيز التي تتحسسهُ هذه الأقطاب تراوحت من 1x10-4) الى 1(1 x 10-M,( 5 x10-4الى1 x10-1)M حد التحسس لهذه الأقطاب كان بحدود (5.5x10-5M (M 8.0 x 10-5, فيما كان معامل الارتباط (0.9997 (0.9998,بالتعاقب.أيضاً تم دراسة تداخلات بعض الأيونات الموجبه مع الأقطاب بواسطة طريقة المحاليل المنفصله وتم تعيين معامل الأنتقائية لهذه الأيونات, وتبين ان تداخلات الأيونات الأحاديه تكون أكبر من تداخلات الأيونات الثنائيه والثلاثية. استخدمت الاقطاب لتقدير الدواء في نماذج قياسية محضرة وكذلك في نماذج دوائية . وان الطرق التحليلية أثبتت أنها طريقة سريعة وبسيطة وتعطي نتائج جيدة.

Abstract

Ion selective electrodes for phenytoin were prepared using sodium tetraphenyl borate (NaTPB) ionophore and various plasticizers: di-n-butylphthalate (DBPH), tri-n-butylphosphate(TBP), o-nitrophenyloctyl ether (ONPOE) and di-octylphenylphosphonate (DOPH). The response characteristics of these electrodes, including slope of the calibration plot, the corresponding concentration range, detection limit, response time, life time, pH effect, and selectivity were studied. The experimental results showed that the best electrode was based on DOPH and DBPH as plasticizers, displaying a linear range from1.00×10−1 M to 1.00 × 10−5 M and 1.00×10−1 M to 2.00×10−5 with a Nernstian slope of 26.6 mV/decade and 26.2 mV/decade, correlation coefficient of 0.9993 and 0.9989, The detection limit was 6.00×10−6 M and 9.00×10−6, the lifetime was around 30 and 28 days respectively. The proposed electrodes were successfully applied to the determination of phenytoin in a pharmaceutical preparation.

الاقطاب الانتقائية لدواء الفينتون تم تحضيرها في هذا البحث من خلال استخدام الليكاند صوديوم تترا فنيل بوريت وتكوين معقد المزدوج الايوني (فينتون-تترافنيل بوريت) ومن ثم تحضير الاغشية البولميرية بوجود بولي فانيل كلورايد وبأستخدام الملدنات الاتية الاتيةdi-n-butylphthalate (DBPH-(1tri-n- butylphosphate(TBP)-2o- nitrophenyloctyl ether (ONPOE)-3di-octylphenylphosphonate (DOPH)-4على التواليوتم دراسة زمن الاستجابة وعمر القطب والدالة الحامضية ووجد ان الاقطاب المحضرة الحاوية على الملدنdi-n-butylphthalate (DBPH-( di-octylphenylphosphonate (DOPH)-تمتلك مى الخطيةfrom1.00×10−1 M to 1.00 × 10−5 M and 1.00×10−1 M to2.00×10−5على التوالي والميل 26.2 and 26.6 ملي فولت/حقبة وعمر القطب 30,28 يوم وعلى التوالي وتم تقدير الأدوية الصيدلانية باستخدام الاغشية المحضرة.

Abstract

A simple, rapid, accurate, precise, specific and economical spectrophotometric method was used for determination of phenytoin .In this study, a second derivative spectrophotometric method was used for the determination of phenytoin (PHT). This method obeys Beer’s law in the employed concentration ranges of 2-10 ppm for phenytoin. The UV derivative spectra 1D for (PHT) have (V) at 233.3 nm,2D spectra for (PHT) have (P) at 226.7 nm, 3D spectra for PHT) have (P) at 223.6 nm, and (V) at 230.4 nm4D spectra for (PHT) have (P) at 236.5 nm, and (V) at 228.5nm.thelinear concentration ranges (2-10) ppm with correlation coefficient (r) 0.9996, 0.9987, 0.9973, 0.9979. The second derivative D2 tech. at (226.7) nm successfully applied for determination the Phenytoin drug in pharmaceutical samples. Results of analysis were validated statistically and by recovery studies.

Abstract

Derivatives of 6-aryl-[5-methyl-1-(4-methoxy phenyl)-1,2,3-triazol-4-yl-s-triazolo[3,4-b]-1,3,4-Thiadiazoles (9) and (10) were synthesized by treating p-anisidine with sodium nitrite followed by the addition of sodium azide. Treating azide compound (3) with ethylacetoacetate yielded 1,2,3-triazolo-4-carboxylic acid (4), which was converted to ester (5) using absolute ethanol. Treating the ester with hydrazine hydrate afforded the acid hydrazide derivative (6), which was reacted with CS2 and KOH to produce the salt potassium dithiocarbazate (7). Addition of hydrazine to the salt produced 1-amino-2-mercapto-5-[5-methyl-1-(4-methoxy phenyl) –1,2,3-triazol-4-yl]-1,3,4-triazole (8). Treating (8) with p-chlorobenzoic acid and anisic acid afforded the target compounds (9) and (10).
