Abstract

BackgroundZygote produce from once a sperm fertilizes an egg cell. Then, the zygote (unicellular) will begin chain of cellular cleavages to produce multicellular mass, its embryo, the differentiated to different tissues and organism.The development of the embryo is called embryogenesis.Coenzyme Q10, is an antioxidant produced in the body. It boosts cellular energy and may enhance the immune system. CoQ10 is present and measurable in seminal fluid,the concentration of CoQ10 directly correlates with both sperm count and motility. It is beneficial inthe prevention and treatment a wide range of health problems.ObjectivesThe present study was aimed to investigate the possibility of using coenzyme Q1O to improvein vitro fertilization (IVF), and early embryonic development (ED) in mice as a model for human beingMethodsSuperovulation program was achieved to mature healthy female mice with age 10-12 weeks and weight 24-26 gm. After sacrificing female, oocytes were collected and incubated within C02 incubator for less than 1 hour. Sperm were collected from vas deference of males. Sperm parameters were assessed after 30 min. of incubation. Mature oocytes were divided intothree groups accordingto theconcentrations of CoQ10 including G1 (control group; SMART medium only), G2 (treated group; SMART medium enriched with 20 M CoQ10) and G3 (treated group; SMART medium enriched with 40 M CoQ10). IVF technique was performed for 3 groups, and assessment of IVF (%), embryonic development stage (%) and abnormal embryo morphology (%) for each embryo stage.ResultsResults of the present study appeared significant increment (P<0.05) in the percentages of IVFfor both treated groups as compared to the control groups.Also, significant increase (P0.05%) in the 8-cells embryo stage were assessed among control and treated groupsConclusionFrom the results of the present study it wasconcluded that the coenzyme Q1O (40µM) enriched to the culture medium improved percentage of in vitro fertilization and no effect on embryonic development.

Abstract

Statin drugs act by inhibiting the enzyme HMG-CoA reductase which is responsible for manufacture of cholesterol and the biosynthesis essential energy production Co-factor (Coenzyme Q10). The aim from this research includes study the effect of statin drugs (Simvastatin and Atorvastatin) on the Coenzyme Q10 using differential pulse polarographic technique at a dropping mercury electrode (DME) and in a mixture of [4:1] methanol-phosphate buffer of pH7.0 as supporting an electrolyte. Prior to this, the behaviors of Simvastatin, Atorvastatin and Coenzyme Q10 were studied separately in their solvents. The half-wave potential (E1/2) of Co Q10 were -0.31volt and -1.37Volt, -1.33 Volt for simvastatin and atorvastatin respectively. A mixture of Coenzyme Q10 with Simvastatin and Atorvastatin in the same solvent shows a shift in their peak potential (Ep) toward more negative potentials values by (-0.06volt) for Simvastatin and by (-0.09volt) by Atorvastatin.

تعمل ادوية الستاتينات على تثبيط انزيم HMG-CoA reductase وهو الأنزيم المسؤول عن تصنيع الكوليسترول والتركيب الحيوي لأنتاج الطاقة Coenzyme Q10. ويتضمن هذا البحث دراسة تآثر ادوية الستاتينات (السيمفاستاتين و الأتروفاستاتين) على مساعد الأنزيم Q10 باستخدام تقنية البولاروغرافيا النبضي المشتق عند قطب الزئبق المتقاطروالمذاب كل منهما في مزيج من الميثانول والمحلول المنظم الفوسفاتي pH7.0 كألكتروليت ساند بنسبة [1:4]. وقبل دراسة التآثر, درس سلوك كل من ادوية الستاتينات ومساعد الأنزيم Q10 بصورة منفصلة في محاليلهما وتم تعيين جهد القمة لمساعد الأنزيم وكانت بقيمة -0.31Voltو بقيمة -1.37Volt, -1.33Volt لكل من السيمفاستاتين والأتروفاستاتين. وتبين ان خليط ادوية الستاتينات مع مساعد الأنزيم Q10 في نفس المذيب ادى الى ازاحة في قيم جهد القمة لمساعد الأنزيم Q10 نحو قيم اكثر سالبية وكانت بمقدار-0.06Volt بالنسبة للسيمفاستاتين و بمقدار -0.09Volt بالنسبة للاتروفاستاتين.

Abstract

Background: Anti-oxidants are used as supplements to counteract the over production of free radicals in periodontaldisease.Co-Q10 functions as an intercellular antioxidant by acting as a primary scavenger of free radicals (FRs) andreactive oxygen species (ROS), this study aimed to evaluate the effect of intra pocket application of perio Q gel(coenzyme Q10) alone and as adjunct to scaling and root planing on the periodontal clinical parameters in thetreatment of patients with chronic periodontitis and compare the better improvement on the clinical periodontalparameters among different treatment modalities at 3 and 6 weeks.Materials and methods: A total of 323 sites with pocket depth (5-8) mm in patients with chronic periodontitis wererandomly divided into three groups. The Gel group, 111 sites were treated with intra-pocket application of Perio Qgel alone. In the Combination group, 106 sites were treated with scaling and root planning (SRP) plus intra- pocketapplication of Perio Q gel, in Scaling and root planing group, 106 sites were treated with scaling and root planingalone. Clinical periodontal parameters such as Plaque index (PI), Gingival index (GI), Bleeding on probing (BOP),Probing pocket depth (PPD), Relative attachment level (RAL) were assessed at first visit, 3weeks and 6weeks.Results: Inter-group analysis, showed significant reduction in the clinical parameters PPD and RAL of combinationgroup in comparison to SRP group.Conclusion: Better improvement of the clinical periodontal parameters had been achieved by using the gel incombination with scaling and root planing instead of using scaling and root planing only

Abstract

Background: Periodontal pathogens can induce free radicals over-formation and thus may cause collagen andperiodontal destruction. Anti-oxidants are used as supplements to counteract the over production of free radicals inperiodontal disease, that can reduce of collagen destruction. Coenzyme Q10 serves as an endogenous antioxidant,regenerates other antioxidants, stimulates cell growth, and inhibits cell death. Because it is an antioxidant, coenzymeQ10 has received much research attention associated with periodontal diseases. Perio Q gel may possibly beeffective as a topical agent and as an adjunct to scaling& root planing in treatment of gingivitis and chronicperiodontitis.Aim of study:Determine the periodontal health status in a follow up study for 6 weeks of a group of patients withchronic periodontitis by measuring clinical periodontal parameters, which included (Plaque index, Gingival index,Bleeding on Probing, Probing Pocket Depth and Relative Attachment Level) and to evaluate the effect of intrapocket application of perio Q gel (coenzyme Q10) alone & as adjunct to scaling,& root planing on the periodontalclinical parameters in the management of patients with chronic periodontitis.Materials and methods: A total of 323 sites with pocket depth (5-8) mm in patients with chronic periodontitis wererandomly divided in three groups. The clinical parameters significantly improved in the phaseof periodontal treatment, indicating that CoQ10 opens new treatment options by improving the host response todisease activity.

Abstract

This study was conducted to investigate the possible protective role of (CoQ10) on reproductive dysfunction of male rats induced by sodium fluoride (NaF). thirty two rats were divided into four even groups and treated for 56 days as follows: the first group served as control (C) The second group (G1) received coenzyme Q10 at dose of 10 mg/Kg.B.W; while third group (G2) received 100ppmsodium fluoride in drinking water and fourth groups (G3) received NaF 100 ppm and CoQ10 10 mg/Kg.B.W for 56 days. The results showed that exposure of rats to NaF caused decrease in serum testosterone (T) and luteinizing hormones (LH) concentrations. Whereas Administration of CoQ10 caused an increase the concentrations of theses hormones in group G3 as compared with group G2. Testicular morphometric and histopathological alterations were observed in group G2 in the form of marked microvacuolation in the basal level of germinal cells, amyloid deposited within seminiferous tubules with disarrangement and depletion of germ cells. Whereas testicular histological alterations were improved in rats treated with CoQ10. In conclusion, the results of the present study showed that use of Co Q10 can alleviating the deleterious effects on male reproductive function following exposure to NaF, may be via improvement the testicular functions or other related endocrine glands.

إن الهدف من هذه الدراسة معرفة الدور الوقائي لمعامل كيو 10 على الخلل الوظيفي في الجهاز التناسلي الذكري في ذكور الجرذان المعاملة بفلوريد الصوديوم. تم استخدم 32 جرذ بالغ قسمت إلى أربعة مجاميع بصورة عشوائية وعوملت لمدة 56 يوماً كالتالي: المجموعة الأولى (س) مجموعه السيطرة أما (ج1) جرعت بمقدار 10 ملغم coQ10 لكل كغم من وزن الجسم، أما المجموعة الثالثة (ج2) جرعت بمقدار 100 جزء بالمليون فلوريد الصوديوم في ماء الشرب، أما المجموعة الرابعة (ج4) فقد أعطيت coQ1010 ملغم لكل كغم من وزن الجسم و100 جزء بالمليون فلوريد الصوديوم في ماء الشرب ولمده 56 يوم. أظهرت النتائج إن التعرض لماده فلوريد الصوديوم سبب تقليل مستوى الهرمون اللوتيني وهرمون الشحمون الخصوي بينما أدى إعطاء ماده الكيو 10 إلى حصول زيادة في تركيز هذه الهرمونات في مجموعة (ج3) مقارنه مع (ج2). لوحظ هنالك تغييرات نسجية في نسيج الخصية لمجوعة (ج2) متمثلة بفجوات صغيره في الغشاء الخلوي للخلايا الجرثومية مع ترسب مادة الامايلود في النبيبات المنوية مع قله تنظيم وعدد الخلايا المشيجية. استنتج من هذه التجربة أن استخدام كيو 10 أدى إلى تقليل التأثيرات الضارة لماده فلوريد الصوديوم على الجهاز التناسلي الذكري فضلا عن تقليل الإجهاد التأكسدي من خلال تحسين وظيفة الخصى أو الغدد ذات العلاقة.

Abstract

Background: Endometrial cancer is the most common gynecologic malignancy in the United States and the fourth most common cancer in women, comprising 6% of female cancers.Objectives: The aim of this study is to investigate the antioxidant vitamins, Coenzyme Q10 and oxidative stress in patients with endometrial cancer.Patients and methods: Fifty six endometrial cancer women patients with various clinical stages (stage 1A, stage1B, stage II, stage III, stage IV) mean aged 58.055 ± 10.561 years, and 30 healthy women volunteers mean aged 39.731 ± 13.504 years, were includes as control group.Results: The results in this study revealed a highly significant decreased (P<0.01) in β- carotene, Vitamin E and significant increased (P<0.01) in (Uric acid and MDA). The results of ( Coenzyme Q10 and Vitamins (A, C) showed a significant decreased (P<0.05), in the sera levels of patients with endometrial cancer as compared to control group.Conclusions: This study suggest that high levels of oxidative stress and low levels of antioxidant defense system may be associated with increased the risks of endometrial cancer.

الخلفية النظرية: سرطان بطانة الرحم هو ورم خبيث خاص بأمراض النساء الأكثر شيوعا في الولايات المتحدة وهو رابع أكثر أنواع السرطانات شيوعا في النساء، يشكل 6% من سرطانات النساء.الغرض من الدراسة: الهدف من هذه الدراسة هو التحقيق من الفيتامينات المضادة للأكسدة، المرافق الانزيمي Q10، والجهد التاكسدي في المرضى الذين يعانون من سرطان بطانة الرحم. المرضى وطريقة البحث: ستة وخمسين من مريضات سرطان بطانة الرحم يتراوح متوسط أعمارهن بين (58،055 ± 10،561 سنة) غير خاضعين للعلاج في هذه الدراسة، و ( عدد= 30) من النساء المتطوعات وهم في حاله صحيه جيده يتراوح متوسط أعمارهن بين (39،731 ± 13،504 سنة)، كمجموعة ضابطة.النتائج: أظهرت النتائج في هذه الدراسة انخفاض بالغ الاهميه (P <0.01) في بيتا كاروتين (ملغرام / ديسيلتر)، وانخفاض معنوي (P <0.01) في فيتامين E (ملغرام / ديسيلتر) وزيادة معنوية (P <0.01 ) في (حمض اليوريك وMDA) ملغرام / ديسيلتر. نتائج (الإنزيم المساعد Q10 وفيتامينات (A، C)) ملغ / ديسيلتر أظهرت انخفاض معنوي (P <0.05)، في مستويات مصل المرضى الذين يعانون من سرطان بطانة الرحم مقارنة بالمجموعة الضابطة.الأستنتاجات: لخصت نتائج هذه الدراسة انخفاض مستويات منظومة الدفاع المضادة للأكسدة نظرا لزيادة الجهد التاكسدي في المرضى الذين يعانون من سرطان بطانة الرحم.
