Cytotoxic effect of crab shell extracts on different tumor cell lines

Abstract

Crab, a kind of marine animal that has a unique status in evolution, has many bioactive substances with special functions. In the present study, we investigated the antitumor effects of four different extracts derived from crab shell (aqueous and methanolic extracts of burned and normal crab shell powder). Aqueous and methanolic extracts of burned crab shell powder were shown to be cytotoxic and inhibit the in vitro proliferation of human laryngeal carcinoma tumor cell lines (Hep-2) and human Rhabdomyosarcoma cell line (RD) and one murine mammary adenocarcinoma tumor cell line (AMN3), in dose and time dependant manner. These results suggest that the burned crab shell powder have an antitumor effect that needs more investigations.

السرطان هو حيوان بحري يمتلك خصائص فريدة في سلم التطور ويمتلك العديد من المواد الفعاله ذات الوضائف الخاصة. في دراستنا الحالية تحققنا من الفعل المضاد للسرطان لاربعة مستخلصات مختلفه مشتقه من قشرة السرطان (المائية والكحولية للقشرة المحروقة والقشرة غير المحروقة). اضهرت المستخلصات المائية والكحولية لمسحوق القشرة المحروقه انها تمتلك سمية خلويه عاليه تجاه الخلايا السرطانية وانها تثبط من تكاثرها ولجميع الخطوط المختبرة (خط سرطان الحنجرة وسرطان الخلية العضلية البشريان وسرطانة الغدة اللبينية الفئري) وان هذا التاثير هو تاثير معتمد على مقدار الجرعه والوقت. هذه النتائج تقترح ان لمستخلصات القشرة المحروقه فعلا مضاد للاورام وهي تحتاج الى المزيد من الدراسات.

Abstract

The leaves of the Capparisspinosa were collected from the eastern Radwaniya/Baghdad duringthe months April, May, June, July, September, and September. The leaves were extracted by water and alcohol.The effectiveness of anti-tumor and anti-leishmania for the water and alcohol extracts of al-Qabar leaf samples for May and September, which were characterized by the highest percentage of active compounds for the rest of the tested months, was evaluated by adopting the results of a previous study for the same researchers or chemical composition and leaf content of active compounds The concentrations of 250, 500, 1000 mg.L1 were evaluated for anti-tumor efficacy. The anti-leishmania efficacy was tested at 250, 500 and 1000 mg L1. The sample of the alcohol extract for May collected complex showed the highest anti-tumor activity at 63.5% at 500 mg.L1and the highest efficacy of the samples of alcohol extract for September was 56.1% at the concentration of 250 mg.L1, while the water extract for the May sample did not show any effectiveness even at the highest concentration 1000 mg L1, but the September collected sample was effective at 49.3% In the case of the inhibitory efficacy of leishmania, the alcohol extract of the May sample did not show on inhibitory activity at the lowest concentration. The effectiveness of 41.3% was observed at 500 mg.L1, but decreased to 14.8% at the highest concentration of 1000 mg L-1. Of the same month at the lowest concentration of 37.8% and decreased effectiveness there was an increase in the anti-leishmania effect of the September extract of the sample with increased concentration. There was of its effectiveness at the lowest concentration of a user and then reached 8 and 37% at the concentration of 500 and 1000 mg.L1 with the same percentage, and the water extract for the September sample did not show any efficacy at the minimum and highest concentrations, except the concentration of 500 mg.L1, which reached 11%.

Cytotoxic effect of esculetin on myeloma and myelogenous leukemic cells.

Abstract

The cytotoxic effect of Esculetin to mouse myeloma and human acute myelogenous leukemia cells invitro was evaluated. Tumor cells exposed to Esculetin in culture showed an exponential cytotoxic responses. A decreased viabilities of both leukemic cells were noticed with 20, 40 and 80 µg / ml Esculetin. Besides, myeloma cells showed a progressive depletion of viability when exposed continuously for 1,2,3 and 4 days, whereas , pretreatment of myeloma cells with Esculetin for 1,2,4 and 24 h followed by removal of the chemical and culturing the cells for 3 days indicated that the time the cells much effected was 24 h post exposure. At this exposure time , exposure to 10 µg / ml decreased the able cell number to 24.66 × 10-4 cells / ml versus 62.66 × 10-4 cells / ml in the control . t – test = 5.16 , p = 0.007 95 % CI = 17.5 – 58.5. Like wise , a 20 µg / ml concentration of esculetin decreased viable cells number to 23.5 × 10-4 cells / ml versus 62.66 × 10-4 cells / ml of control. T-test 7.39 , P = 0.002. 95 % CI = 24.5 – 53.8. The efficiency of Esculetin in protecting mice implanted with syngeneic myeloma was studied . The results of invivo experiment demonstrated that the chemical could protect mice from lethal dose of the experimentally implanted tumor . The protection value reached 66.7 % in animals treated with 200 or 400 µg Esculetin for 10 consecutive days. For 200µg drug plus myeloma versus myeloma only , chi – square value was 3.086 p=0.08 and 95% CI = 0-2.4. However , at 400 µg drug plus myeloma versus myeloma only , the chi – square analysis showed a value of 4.17 , P=0.04 95% CI = 0.02 – 1.3. Besides , the survival of the animals was increased . The therapeutic relevance and speculated mechanism of Escletin cytotoxic effects are discussed. In conclusion , it seems that esculetin has antiproliferative effects to mouse myeloma and to human myelogenous leukemia cells.

تم تقدير التأثير المسمم لخلايا الاسكولتين تجاه خلايا الفأر المايلومية والخلايا السرطانية النقوية العائدة للانسان. اظهرت الخلايا المعرضة للاسكولتين استجابة تسممية تصاعدية. اظهرت الجرع 20 و 40 و 80 مايكروغرام / سم3 تأثيرا في انخفاض الحيوية لكلا النوعين من الخلايا السرطانية. اضافة الى ذلك فان خلايا المايلوما اظهرت نقصانا مستمرا في الحيوية عند التعرض ليوم او ثلاثة او اربعة ايام. وقد كان بدء التاثير في الاربع والعشرين ساعة بعد التعرض. اذ كان في هذا الوقت وبتعرض 10 مايكروغرام/سم3 عددا يقدر بـ 104 × 24.66 خلية / سم3 للخلايا المعرضة مقارنة ب 104 × 62.66 للخلايا غير المعرضة لمادة الاسكولتين..( t = 5.16 , P = 0.007 , 95 % CI = 17.5 – 85.5 ) كما ان التعرض لجرعة 20 مايكروغرام / سم3 ادى الى نتائج مقاربة. تمت دراسة تاثير الاسكولتين في حفظ الفئران المستلمة لخلايا المايلوما . اشارت النتائج الى ان الاسكولتين له تاثير حافظ لتلك الفئران.اذ بلغت نسبة الحفظ 66.7 % للحيوانات المعاملة بجرع 200 او 400 اسكولتين لمدة 10 ايام متتالية واشارت نتائج التحليل الاحصائي ان النتائج ذات شأن للجرعتين. تم مناقشة امكانية المادة العلاجية والية عمل المادة المسممة

Abstract

Mushroom is widely used as a traditional medicinal fungus, and it has been known to exhibit immune-stimulatory and anti-cancer activity. However, the exact immune response in tumor-bearing mice after exposure to mushroom extract was unclear. The aim of this study was to investigate whether or not crude preparations extracted from Agaricusbisporus can promote type 1 immune response in tumor-bearing mice.Watery and ethanolic extracts of mushroom were intraperitoneally administered into tumor-bearing mice for 28alternative days, tumor volume was measured twice weekly, also IL-2 level was determined in the sera of mice oneday before the beginning of treatment and after 2 and 4 weeks of treatment. The results showed that both extractssignificantly inhibit the growth of tumor up to 72.2% at the end of experiment, while only the ethanolic extractsignificantly increased the level of IL-2 production from 11.2 and 16.9 pg/ml up to 26.4 and 42.5 pg/ml after 2 and 4 weeks of treatment respectively.To confirm these results , further studies are required to investigate the ability of these extracts to promote the production of IL-12 & IFN-γ which are the major cytokines involved in immune response type 1 activation.
