Abstract

The study concerned with describing the activity and some properties of the enzyme inorganic pyrophosphatase (EC 3.6.1.1) from Escherichia coli. A high activity of the enzyme was detected in Escherichia coli. The highest specific activity of the enzyme was detected in the supernatant (466) nmol/min/mg protein and the lowest was in the precipitant, by a sonication and centrifugation, (98) nmol/min/mg protein, while crude extract exhibited a value between the two limits (420) nmol/min/mg protein. The optimum conditions of the enzyme activity were (300) mg protein as a source of enzyme, (2.0) mmol of sodium pyrophosphate as a substrate, at pH (7.0) , with (10) minutes as incubation time at 37C and the Michaelis-Menten constant (Km) was about (0.42) mmol.

تضمن البحث دراسة فعالية وبعض الصفات الحركية لانزيم البايروفوسفاتيز اللاعضوي inorganic pyrophosphatase (EC 3.6.1.1) المستخلص من جرثومة القولون Escherichia coli ، اظهرت النتائج فعالية عالية لهذا الانزيم في هذا النوع من الجراثيم ، وكانت اعلى فعالية نوعية تم الحصول عليها هي (466) نانومول / دقيقة/ ملغم بروتين في رائق supernatant خلايا جرثومة القولون. كانت الفعالية النوعية اقل في المستخلص الخام crude extract لخلايا جرثومة القولون ، اذ بلغت (420) نانومول/دقيقة/ملغم بروتين، في حين اظهر الراسب precipitant المرسب بطريقة الموجات فوق الصوتية والطرد المركزي اقل فعالية نوعية للانزيم اذ بلغت (98) نانومول /دقيقة/ملغم بروتين . وجرى تحديد الظروف المثلى لفعالية الانزيم عند (7.0= pH) وكانت اعلى فعالية نوعية للانزيم عند الظروف (300) مايكروغرام من بروتين رائق الخلايا كمصدر للانزيم و (2.0) ملي مول من المادة الاساس بايروفوسفات الصوديوم Na4ppi وفترة حضن لمدة (10) دقائق عند الدرجة الحرارية (37)م ، وبلغت قيمة ثابت ميكيلس – منتن (Km) Michaelis-Menten مقاربة الى (0.42) ملي مول .

Abstract

Marsh land ( south of Iraq ) is of great international interest at present time. Studying some microbial properties including enzyme activities , can be useful as indicator of soil quality of this area. General conclusion of enzyme reaction in soil can be reached from the thermodynamic parameters of the enzyme – soil system. Hence , this experiment was conducted to study thermodynamic parameters ( Q10 , Ea , ΔHa , ΔSa and ΔGa ) for inorganic pyrophosphatase enzyme in this soil as compared with their counter parts of other soils of south of Iraq ( Al-Zubair and Abul-Khasib soils ). Results indicated that Q10 and ΔGa values of marsh land did not significantly differs from other soils under study with average values of about 1.11 and 14.55 KJmol -1 ,respectively. Ea values of marsh land , Al-Zubair and Abul-Khasib soil were 8.72 , 7.47 and 9.53 KJmol -1 ,respectively. ΔSa values for marsh land soil(–10.49 J deg -1 mol -1 ) were lower than those of Abul-Khasib ( –7.85 J deg -1 mol -1) but higher than those of Al- Zubair (–14.55J deg -1 mol -1 ).ΔHa values were in order of Abul-Khasib ( 12.08 KJmol -1 ) > marsh land ( 11.27 KJmol -1 ) > Al-Zubairsoil ( 10.02 KJmol -1 )

Abstract

Marsh land (south of Iraq) is of great interest at present time, therefore studying some of microbial properties, including enzyme activities, can be useful as indicator of soil quality of this area. Hence, this study was carried out to compare inorganic pyrophosphatase (EC 3. 6. 1. 1) activity in this soil with their counterparts of other soils of south Iraq (Al–Zubair and Abul–Khsib soils). Results indicated that inorganic pyrophosphatase activity at all substrate concentrations in marsh land soil was higher than those of Al–Zubair soil, but lower than those of Abul–Khasib soil. Expressed as PO4-3 – P released per gram soil per 5 hrs, the average Vmax values were 21.93, 37.23, and 33.33 while average Km values were 19.82, 18.26 and 29.77 for Al–Zubair, Abul–Khasib and marsh land soils, respectively. The results also revealed that the threshold of salinity negative effect on inorganic pyrophosphatase activity varies according to initial E.C. of soils (6 and 12 dS.m-1 for Abul–Khasib andmarsh land soils, respectively).
