Coronal sealing ability of different sealers on teeth obturation and the effectiveness of two different obturation techniques

Abstract

Background and objectives: The purpose of this study was to evaluate the effect of two sealers on coronal seal and the coronal sealing ability of two obturation techniques. Methods: Forty extracted single–rooted human teeth were instrumented with the step-back technique and were irrigated with 5.25% sodium hypochlorite (NaOCl). The smear layer was removed by washing with 10 ml of 17% ethylene diamine tetra-acetic acid (EDTA) and 10 ml of 5.25% (NaOCl). All instrumented root canals were randomly divided into two experimental groups; Group A: 20 roots epoxy resin sealer (AH26) was used, Group B: 20 roots Zinc oxide euginol (ZOE) sealer was used. Each group of main groups were subdivided randomly into two obturation groups; 1. 10 roots obturated by thermo plasticized technique (obtura II), 2. 10 roots obturated by cold lateral compaction technique. The root surfaces were then coated with nail polish except for the coronal, then dye penetration study was conducted and samples were examined under the stereomicroscope. Data had been collected from three independent examiners and statistically analyzed using student t-test. Results: There were significant differences between AH26 with ZOE in group obturated by obtura II and when obtura II technique compared with the cold lateral compaction technique in group of AH26 sealer. Conclusion: Epoxy resin sealer (AH26) and obtura II gave the best results in coronal sealing when compared with Zinc oxide euginol sealer and cold lateral compaction technique.

In vitro evaluation of the Antimicrobial Activity of four resin based Endodontic Sealers on three bacterial species

Abstract

Aim: The aim of this in vitro study was to evaluate the antimicrobial action of fourresin based endodontic sealers after 24 hrs.Materials and Method: The freshly mixed sealers were AH26, AH-plus, Epiphanyand Topseal. They were prepared according to manufacturer’s instruction andplaced in prepared wells of 40 agar plates inoculated with Streptococcus viridans,Staphylococcus aureus and Enterococcus faecalis. (10 samples for eachmicroorganism). Four cavities, each one measuring 5 ml in diameter and 4 ml indepth, were made in each agar plate using cork poorer. Agar diffusion method onMuller Hinton agar was employed, and zones of inhibition were measured after1day.Results: AH26 containing proved to be the most effective against all microorganismstested. This was followed by Epiphany. Topseal which showed antibacterialactivity on all tested microorganisms slightly higher than that of AH-Plus whichshowed the least action on all tested microorganisms.Conclusion: All the sealers evaluated in this study showed different inhibitory effectagainst all bacterial strains.

Abstract

Background: One of the major problems in endodontics is micro-leakage of root canal fillings which might contribute to the failure of endodontic treatment. To avoid this problem, a variety of sealers have been tested. The objective of this, in vitro, study was to evaluate the shear bond strength of four resin based sealers (AH plus, silver free AH26, RealSeal SE and Perma Evolution permanent root canal filling material) to dentin.Materials and Methods: Forty non-carious extracted lower premolars were used. The 2mm of the occlusal surfaces of teeth were sectioned, to expose the dentin surface. The exposed dentin surfaces of teeth were washed with 5ml of 2.5% NaOCl solution followed by 5ml of 17 % EDTA then rinsed by deionized water to remove the smear layer. The teeth were divided into four groups according to the type of sealer used: Group A: silver free AH26, Group B: AH plus., Group C: RealSeal SE, Group D: Perma Evolution. Polyethylene tube cylinders (4mm internal diameter & 5mm length) were fixed on the dentin surfaces. Then freshly mixed sealers were poured into the tubes and allowed to bench set for two hours and were stored at 100% humidity and 37˚C for one week. With an Instron machine, the shear bond strength between the tested sealers and the dentin, in Mpa, was measured. Statistical analysis was carried out using the one-way ANOVA and Student’s t-tests.Results: Perma Evolution scored the highest mean value of sheer bond strength, being 3.343 Mpa followed by the AH plus (2.786 Mpa) and AH26 (2.149 Mpa). While the RealSeal scored the lowest mean value of sheer bond strength, which was (1.831 Mpa). ANOVA test results showed a highly statistically significant difference. Student's t test results revealed significant differences between all the compared groups, except one paired group had a non-significant difference in the shear bond strength which was between the AH plus and Perma Evolution groups (P>0.05).Conclusions: The results of this study pointed to Perma Evolution which scored the highest sheer bond strength between the tested sealers

Abstract

Background: The bond strength of root canal sealers to dentin and gutta-percha seems to be an important propertyfor maintaining the stability of root canal filling, which potentially influences both leakage and root strength. Theobjective of this, in vitro, study was to evaluate the shear bond strength of three different endodontic sealers (Gutta-Flow, AH Plus, Apexit Plus) to dentin, in the presence and absence of the smear layer and gutta percha.Material and Methods: After slicing off the occlusal 2mm of 60 extracted human maxillary premolar teeth, theexposed dentin served as the tested surfaces; the teeth were fixed with cold cure acrylic, and were divided into twogroups according to the smear layer presence, group A without smear layer, when dentin surfaces were irrigatedwith EDTA 17% followed by distilled water then subdivided into 3 subgroups according to the type of sealer used;group B when dentin surfaces were washed with distilled water only, then subdivided into 3 subgroups. Thirty samplesof gutta-percha were prepared and named as group C which was subdivided into 3 subgroups. Five mm longsection of polyethylene tubes were placed on the dentin or gutta percha surfaces and filled with freshly mixedsealer. After one week, all the samples were tested for shear bond strength by the Instron Universal Testing Machineat a cross head speed of 0.5 mm/min. The data was calculated in MPa and was statistically analyzedResult: There was a highly significant difference in the shear bond strength (P < 0.05) in comparison among the testedgroups, GuttaFlow showed non-significant difference in bond strength to dentin with and without smear layer, whileAH Plus and Apexit Plus showed a high significant difference.Conclusions: AHPlus showed the highest shear bond strength in all the tested samples, while GuttaFlow was the least.Additionally, AH Plus and Apexit Plus shear bond strengths were affected by the smear layer removal, whileGuttaFlow was not.

Gutta-percha أن لقوة الرابطة لسدادات قناة الجذرالى عاج السن وو عاج السن بوجود اوعدم (AH Plus, Gutta-FlowApexit Plus,) كان الغرض من ھذه الدراسة تقییم قوة الترابط القصي بین ثلاثة انواع من السدادات اللبیة المختلفةبعد قطع 2ملمیتر من تاج 60 سن من أسنان الإنسان(الضواحك العلیا) . تم تثبیت الأسنان في مادة الاكریلیك ، و قسمت .Gutta-percha و ال Smear layer وجود طبقة ال.Smear layer الى مجموعتین وفقا لوجود طبقة ال(AP-) %17 یلیھ الماء المقطر وقسمت إلى 3مجموعات فرعیة وفقا لنوع السدادة المستعملة EDTA على سطح عاج السن مع غسل بمادة Smear layer المجموعة (أ) بدون.D, AH-D, GF-D. APD-S, AHD-S, GFD-S)( المجموعة (ب) غسل سطح عاج السن بالماء المقطر فقط ثم قسمت إلى 3 مجموعات فرعي.(AP-G, AH-G, GF-G) أعدت لتكون المجموعة (ج)وقسمت إلى 3 مجموعات فرعیة (Gutta-percha) ثلاثونعینة مناختبار قوة الرابط القصي تم بعد اسبوع . . Gutta-percha استعمل انبوب بطول 5 ملم من البولي اثیلین بسداده مخلوطة حدیثا ووضع على سطح عاج السن أو السجل فرق معنوي غیر ملحوظ في قوة الترابط مع عاج السن بوجود Gutta-Flow. تم حساب البیانات بوحدة المیغا باسكال و تم تحلیلھا إحصائیاً . أظھرت النتائج فرقا معنویا كبیراسجلوا فرقاً معنویاً عالیاً. Apexit Plus و AH Plus بینما Smear layer اوعدم وجود التأثرت Apexit plus و AH Plus الاقل بینھم . وقوة الربط القصي لل Gutta-Flow اظھر اعلى قوة ربط قصي في جمیع العینات بینما كان AH Plus : الاستنتاج النھائيفلم تتأثر.

Abstract

The present study aimed to evaluate the antimicrobial properties of aqueous ginger extract as endodontic sealer, against Staphylococcus aureus, Enterococcus faecalis, Streptococcus sanguis, Candida albicans, anaerobic bacteria and normal flora of oral cavity, and to evaluate the most effective dilution of ginger extract against Enterococcus faecalis in dentinal tubules in order to use it as one component of root canal sealer. Disc diffusion test and direct contact method were used to evaluate the antimicrobial properties of the aqueous ginger extract. The antimicrobial activity was tested 1, 3 and 30 days after dentinal tubules manipulation using different dentin sealer. The Results showed that the highest antimicrobial activity was exhibited by the 20% (w/v) aqueous ginger extract. Moreover, this ginger extract (20%) showed a remarkable antibacterial activity against Enterococcus faecalis in infected dentinal tubules when examined in vitro; the study indicates that ginger extract might have promising effect to be use as one component of root canal sealer.

Abstract

Background: The bond strength of the root canal sealers to dentin is very important property for maintaining the integrity and the seal of root canal filling. The aim of this study was to evaluate and compare the push-out bond strength of root filled with total fill Bioceramic, AH Plus and Gutta-flow®2 sealers using GuttaFusion®obturation system versus single cone obturation technique.Materials and method: sixty of mandibular premolars teeth with straight roots were used in this study, these roots were instrumented using Reciproc system, instrumentation were done with copious irrigation of 3 mL 5.25% sodium hypochlorite solution (NaOCl) during all the steps of preparation, and smear layer will be removed with 1 ml of 17% EDTA kept in the canal for 1 min, roots were randomly divided into two groups according to the obturation technique (thirty teeth for each group):Group I: Single Reciproc Gutta percha cone obturation technique, Group II: Gutta fusion obturation technique, then each group divided into three subgroup according to the type of sealer, AH subgroup: AH Plus sealer, BC subgroup: bioceramic sealer and GF subgroup: Gutta flow 2 sealer. The roots then stored in moist environment at 37°C for one week, the roots were embedded in clear acrylic resin and each root sectioned into three levels apical, middle and cervical. The bond strength was measured using computerized universal testing machine each section fixed in the machine so that the load applied from apical to cervical direction at 0.5mm/min. speed and the computer show the higher bond force before dislodgment of the filling material. These forces were divided by the surface area to obtain the bond strength in MPa.Results: Statistical analysis was performed and the result showed a highly significant differences between the three types of sealers when the same obturation technique were used, also there is highly significant differences between two groups with two different obturation technique.Conclusion: This study showed that the push out bond strength of AH plus sealer was higher than bioceramic sealer and Gutta flow 2 sealer respectively when the same obturation technique was used. The push out bond strength was affected by the obturation technique and Gutta fusion obturation technique showed higher bond strength than single cone obturation technique when the same type of sealer was used

Abstract

Aims: The aims of present study were to compare shear bond strength (SBS) and push-out bond strength (PBS) of three types of sealers to gutta percha (G) and dentin (D) and to assess failure modes at debonded interfaces. Materials and Methods: Sealers tested were AH Plus, Sealer 26, and Endofill. In PBS, 30 mandibular premolar decoronated and instrumented with rotary ProTaper to F3 then teeth were divided into 3 groups (10 teeth in each) and obturated with F3 proTaper G and one of tested sealer, 1mm mid root slices were prepared for PBS test. In SBS/D, 30 root cylinders 7 mm length were embedded horizontally in resin then root surfaces were flattened and smooth. In SBS/G, 30 G cylinders were embedded in a resin. In both SBS tests, specimens were divided into three groups, 10 samples each then polyethylene tubes positioned on polished specimens (prepared for SBS/D and SBS/G) filled with one of the tested sealer. All BS tests were performed on digital universal testing machine. Collected data analyzed statistically using one way ANOVA and Tukey test. Results: PBS, SBS/D, and SBS/G for AH Plus significantly higher than Sealer 26 and Endofill. Also Sealer 26 exhibited significantly higher bond strength than Endofill. Predominance mode of failures were adhesive and mixed. Conclusions: According to the finding of present study, it can be concluded that the AH Plus for root canal therapy produce better results in endodontic. PBS and SBS were sufficient tests to measure bond strength of root canal sealers to G and D.

Abstract

Background: Antibacterial action of root canal filling is an important factor for successful root canal treatment, so theaim of the study was to identify and to compare the antimicrobial effect of new sealer (GuttaFlow) to commonlyused endodontic sealers (AH Plus, Apexit and EndoFill) against four endodontic microbes.Materials and methods: Twenty patients aged (30-40) years with infected root canals were selected. Four types ofmicroorganisms were isolated from root canals (E faecalis, Staphylococcus aureus, E coli and Candida albicans) andcultured on Mueller Hinton agar Petri-dishes. After identification and isolation of bacterial species, agar diffusionmethod was used to assess the antibacterial action of four contemporary endodontic sealers used in root canalobturation (AH Plus, Apexit, EndoFill and GuttafFlow). Four wells measuring (5mm depth and 4mm diameter) werecreated in each Petri dish and sealer was applied into them incubated overnight at 37 C° for bacterial species and48 hr. at 37 C° for Candida albicans prior to determination of results. Zones of inhibition (no growth of bacteria) wereexamined around the wells containing sealer & diameters of the zones were measured in mm. The mean of inhibitionzones for each group was measured and statistically analyzed among groups using ANOVA and between groupsusing LSD tests.Results: There was a highly significant difference (P<0.001) among all the tested groups. EndoFill showed themaximum antibacterial action against tested microorganisms. GuttaFlow showed moderate to weak antimicrobialeffect, Apexit had weak effect, while AH Plus had no antibacterial action.Conclusion: All the tested materials except AH Plus had antibacterial efficacy against E faecalis, Staphylococcusaureus, E coli and Candida albicans. EndoFill had favorable results among tested sealers and E faecalis was the mostresistant bacteria, but none of the materials totally inhibited microbial growth. Thus, endodontic treatment must beperformed under aseptic conditions

Abstract

The aim of this study was to evaluate the apical sealing ability of GuttaFlow2 obturation material using dye penetration method. Palatal roots of forty five extracted maxillary first molars were instrumented with ProTaper rotary files to size F4. The teeth were divided into three experimental groups according to type of sealer used, group1: root obturated with AH Plus sealer and single cone technique, group2: root obturated with GuttaFlow2 sealer and single cone technique, group3: root obturated with GuttaFlow2 sealer only. Results: Data was subjected to statistical analysis. P-Value<0.05 was considered as significant. The results of this study showed that Gutta-Percha/AH Plus showed the lowest mean of leakage with no statistically significant difference with group2 and statistically difference between Gutta-Percha/AH Plus and group3.Conclusion: The use of GuttaFlow2 with a single gutta-percha master cone creates an apical seal that is equivalent to that produced with gutta-percha/AH Plus sealer using single cone technique, GuttaFlow2 when used as only creates a poorer sealing than when used with single cone techniques.

ان الهدف من هذه الدراسة هولتقييم الختم القمي لسدادة القناة GuttaFlow2 باستخدام طريقة التسرب الصبغي الدقيق. تم اختيار خمسة وأربعين رحى اولى علوية وتم تحضيرها باستخدام ادوات تحضير دوارة نوع بروتيبر حجم F4. تم تقسيم الأسنان إلى ثلاث مجموعات تجريبية وفقا لنوع الماد اللاصقة، المجموعه الاولى: جذور محشاة بمادة Plus AH باستخدام طريقة تقنية المخروط الواحد، المجموعه الثانية: جذور محشاة بمادة GuttaFlow2 باستخدام طريقة تقنيةالمخروط الواحد، المجموعة الثالثة: جذور محشاة بمادة GuttaFlow2 سداده فقط.النتائج: البيانات تعرضت للتحليل الإحصائي , اذا كانت قيمه P <0.05 فيدل على وجود فرق معنوي. وأظهرت نتائج هذه الدراسة أن الكوتابيرشا /Plus AH أظهر أدنى مستوى تسرب ذروي مع عدم وجود فروق ذات دلالة إحصائية مع المجموعه الثانية مع وجود فرق معنوي بين الكوتابيرشا / Plus AH و المجموعة الثالثة.الخلاصة: إن استخدام GuttaFlow2 مع كوتابيرشا مفردة ينتج عنه ختم قمي يعادل الختم القمي التي أنتجت مع الكوتابيرشا / Plus AH باستخدام تقنية المخروط الواحد، GuttaFlow2 عندما يستخدم لوحده ينتج عنه ختم قمي ضعيف مقارنة عند استخدامها مع تقنية المخروط الواحد.

Abstract

الهدف من هذه الدراسة المختبرية كان لتقييم قدرة إحكام قمة الجذر لثلاثة حشوات جذور طلائية مختلفة باستخدام طريقة التخلل الصبغية. خمسة وأربعون جذر حنكي لأسنان مقلوعة وهي الضرس الأول من الفك العلوي تم تحضيرها بمبارد الحفر الآلي (بروتابر) إلى الحجم ف4.قسمت هذه الأسنان إلى ثلاثة مجموعات مختبرية وفقا لنوع حشوة الجذر الطلائية المستخدمة, حيث كانت المجموعة الأولى تمثل الجذور التي تم حشوها باستخدام (اي اتش بلاس), و المجموعة الثانية تمثل الجذور التي تم حشوها باستخدام (اكتف جي بي), والمجموعة الثالثة للجذور التي تم حشوها باستخدام (جوتا فلو 2) وكانت النتيجة أن (اي اتش بلاس) قد اظهر اقل معدل للتسرب مقارنة بكل من ( اكتف جي بي ) و ( جوتا فلو2).
