Digital Image Analysis versus Visual Scoring for Bcl-2 and P53 Protein Expression in Glioma

Abstract

Background: Traditionally, evaluation of the results of immunohistochemistry was done by visual quantification. Materials and methods: for reliable evaluation, more time-efficient and user friendly method we used simple computer program with image analysis options as independent parameters for reading positive results. To test the validity of visually scored results, we compare and correlate the results of Digital image analysis (DIA) variables with the visual scores of 280 pictures taken from entire stained glioma tumor sections for Bcl-2 and P53 oncoproteins in different glioma tumor grades.
Results: In this study, rates expression of both oncoproteins was evaluated visually in glioma tumor samples (Bcl-2=72.41% and P53=82.76%), no statistical significant differences were observed according to pathologic grades. Similarly, these results were also explained by data obtained by DIA variables that has been closely correlated with visual scores. More importantly, the DIA variables explained little discrepancy in the visual scores of both oncoproteins. Conclusions: the quantitative DIA measurements of the immunohistochemically stained sections made the results more objective and supported pathomorphological diagnosis.

Abstract

Background: Coated aesthetic archwires are currently the existing solutions for the esthetic problem, but the color ofthese archwires tends to change overtime. This study was aimed to evaluate the color stability of different types ofesthetic archwires from four different companies at different time periods after immersion of two different stainingdrinks which are tea and Pepsi cola.Materials and methods: 48 specimens were prepared, each specimen contains 10 aesthetic archwires; and weredivided according to type of solution into two groups: A (tea) &B (Pepsi cola); each group contained 24 specimens(12 specimens were immersed in the staining solution as 4 specimens for each time intervals and 12 specimens wereimmersed in distilled water as control group). Each subgroup contains one specimen from each company. Colormeasurements were performed by means of a computed spectrophotometer according to the CommissionInternationale de I’Eclairage L*a*b* system, and color changes (ΔE*) and National Bureau of Standards units.Results: Both staining solution caused color change in variable degree and the higher amount of color instability wasfound for the archwires from the Hubit Company while the least amount of color change was for the archwires fromthe Orthotechnology Company.Conclusion: The tea caused highly significant change on the aesthetic archwires which is higher than the effect ofthe Pepsi cola

Abstract

Bladder cancer is the ninth most common malignancy all over the world. Immune picture is important in predicting clinical outcome with bladder cancer, so the aim of the present study was to investigate the number of killer cells in the tissue of the patients with the malignant and benign bladder cancer before treatment by using an antigen retrieval immunohistochemical method on archived bladder tumors tissue. In our study, the number of the patients tissue with malignant cancer that staining by immunohistochemistry of NK cells (20%) was lower than in benign (80%) (p≤0.01). The results of the present study were indicated that the lower level of the NK cells in the malignant tissues may be leads to impaired anti-tumor immune response toward the tumor cells and is subsequently unable to suppress progression of the tumor.

يأتي سرطان المثانه بألمرتبه التاسعه من حيث الانتشار عالميا , حيث ان الصوره المناعيه لمرضى سرطان المثانه هامه في ما يتعلق بالواقع السريري للمرضى وتطوره ولهذا هدفت الدراسه الحاليه الى التحري عن اعداد الخلايا القاتلات الطبيعية في انسجة مرضى سرطان المثانه الخبيث والحميد قبل العلاج. وقد بينت نتائج الدراسه الحاليه 23.3% من الانسجة السرطانيه من النوع الخبيث فقط ايجابيتها الى الصبغه الخاصه بخلايا القاتلات الطبيعيه بينما كانت النسبه الاكبر 80%الى الانسجه من النوع الحميد . استنتج من الدراسه الحاليه ان النسبه المئويه القليله لخلايا القاتلات الطبيعيه في انسجة السرطان من النوع الخبيث تعكس ضعف المناعه الطبيعيه في المرضى وقد تكون السبب في نشوء وتطور سرطان المثانه .

Abstract

Background: The demand for better esthetic during orthodontic treatment has increased nowadays, so orthodontistsstarting using esthetic arch wires, brackets and ligatures.Tooth colored brackets were introduced in different types ofmaterials. Sapphire ceramic brackets are one type of esthetic brackets and their color stability remains the mainconcern for the clinicians and patients at the same time .The present study design to evaluate the effect of three different staining materials (pepsi, black tea and cigarettesmoke) on the stainability of sapphire ceramic brackets bonded with three types of light cure orthodontic adhesiveswhich include: Resilience, Enlight and Transbond.Materials and Methods: The sample consisted of three hundred sixty sapphire brackets. The brackets were dividedaccording to bonding materials into three groups each group consist of one hundred twenty brackets, then eachsubgroup farther subdivided into four groups according to the material they were immersed (distilled water, blacktea, Pepsi and cigarette smoke) with thirty brackets each, then Each group with ten brackets farther subdividedaccording to time interval of immersion in each media into three groups one day, seven days and fourteen days at37°C in the incubator.A UV-Visible spectrophotometer (Shimadzu, UV -1800) was used to perform a light absorptiontest.Results: ANOVA and LSD post Hoc tests were used to identify the significant effects of the staining materials at asignificance level P ≤ 0.05.It was found that the immersion time gradually influenced the color stability of the adhesivematerials with sapphire brackets with the highest activity observed at fourteen days interval. The brackets bondedwith Resilience light cure adhesive are the most type affected by staining materials, then followed by the bracketsbonded with Transbond and finally the brackets bonded with Enlight light cure adhesive. For the staining materials itwas found that the cigarette smoke is the most powerful staining material, followed by tea and finally pepsi.Conclusions: From the above result we can conclude that the type of adhesive must take in consideration when theesthetic brackets have been used

Abstract

Aims: This study investigated the effect of polishing times (immediately, 1 hour and 24 hour after light curing) and staining solutions on the color stability of two composite resins. Materials and Methods: Nanocomposite (Filtek Z350) and microhybrid composite (Valux plus) were used. Sixty discs (8mm diameter×2mm height) were made for each composite, and divided into four groups: the first group was polished immediately after light curing; second group was polished after 1 h; third group was polished after 24 h of curing; and the fourth group was the control. Color was measured with spectrophotometer. After baseline color measurements, the specimens of each group divided into three subgroups; specimens of first subgroup were immersed in coffee; specimens of second subgroup were immersed in orange juice; specimens of third subgroup were immersed in distilled water, after 7 day of immersion, the color was measured again and ΔE were calculated. Statistical analysis: Two-way ANOVA, Duncan's Multiple Range test and t-test. Results: The influence of polishing times and staining solutions on color change was significant. Polishing 24 hours after curing presented lowest ΔE value. Coffee promoted more color change than orange juice and distilled water, Filtek Z350 showed lower ΔE than Valux plus. Conclusions: Immediate polishing negatively affect the color stability of composite resins, coffee had a significant influence on discoloration of composite resins. The nanocomposite showed higher color stability than microhybrid composite.

Abstract

The study included the relying of some phenotypic tests to investigate the possibility of isolation and identification of oral Treponema from infected periodontal pocket samples. These phenotypic tests included the examination of fixed smears prepared from the bacterial isolates and staining them with gram and modified gram's stain and with Fontana stain under light microscope where the distinct spiral forms of germs were seen. The oral Treponemes also identified by the observation of their actively motile spiral cells in wet smears using phase contrast microscope. The isolation and identification of these bacteria also proved after their interaction with specific antibody and cell clarifying by greenish spiral form when illuminated with U.V. light using fluorescence microscope. Further, the isolation and identification of oral Treponema was continued by the removal of their outer membrane (O.M.) using Triton X-100 and releasing their flagella and staining with specific flagella stain.

تضمنت الدراسة اعتماد بعض الاختبارات التشخيصية المظهرية للتحري عن امكانية عزل جراثيم Treponema الفم من عينات جيوب ما حول الاسنان المصابة، شملت هذه الاختبارات فحص مسحات مثبتة محضرة من العزلات الجرثومية وصبغها بصبغة كرام وكرام المحورة وصبغة فونتانا تحت المجهر الضوئي حيث لوحظت الاشكال الحلزونية المميزة للجراثيم. كما شخصت Treponema الفم بملاحظة خلاياها الحلزونية المتحركة بنشاط في المسحات الرطبة باستخدام مجهر الطور المتباين. هذا وتم تأكيد عزل وتشخيص هذه الجراثيم بعد مفاعلتها مع الاجسام المضادة المتخصصة ووضوح خلاياها الحلزونية باللون الأخضر البراق عند تسليط ضوء UV عليها باستخدام المجهر المتألق، اضافة الى اننا تمكنا من ازالة الغلاف الخارجي (O.M.) باستخدام مادة Triton X-100 وتحرير اسواطها وصبغها بصبغة السوط الخاصة.

Abstract

The present study was evaluated the anticancer activity of Radix Pulsatillae ethanol extract at first by using high throughput screening HTS for herb fractions screening in concentration of 100μM to identify the active fraction and then we measured the cytotoxic effects by using morphology changes, and detected apoptosis by Hoechst 33258 staining. Our results showed that Radix Pulsatillae ethanol extract has a strong toxicity against A549 lung cancer cells and low toxicity with mice normal splenocytes and requires more investigation to identify the small natural molecule with new anticancer activity and further mechanistic for cancer pathways.

حضر المستخلص الايثانولي لنبات رادكس بولساتيللا باستخدام تقنية الحقن الانتاجي العالي الجريان. ولتحديد الفعالية السرطانية ضد خلايا سرطان الرئة تم قياس السمية عن طريق تجربة تأثير المستخلص قبل وبعد الاضافة وبتركيز 100 مايكرومولار على تغير شكل وطبيعة الخلايا السرطانية وكذلك تجربة كشف برنامج الخلايا المميت. دلت النتائج ان لمستخلص الايثانول لرادكس بولساتيللا فعالية قوية ضد سرطان الرئة واقل فعالية ضد خلايا طحال الفئران الطبيعية حيث يحتاج الى دراسة اعمق من اجل عزل وتشخيص المركب المسؤول عن هذه الفعالية المضادة لخلايا سرطان الرئة ودراسة الميكانيكية الفعالة لدورات الخلايا السرطانية

Abstract

Background: One of the drawbacks of vital teeth bleaching is color stability. The aim of the present study was toevaluate the effects of tea and tomato sauce on the color stability of bleached enamel in association with theapplication of MI Paste Plus (CPP-ACPF).Materials and Methods: Sixty enamel samples were bleached with 10% carbamide peroxide for two weeks thendivided into three groups (A, B and C) of 20 samples each. After bleaching, the samples of each group weresubdivided into two subgroups (n=10). While subgroups A1, B1 and C1 were kept in distilled water, A2, B2, and C2were treated with MI Paste Plus. Then, the samples were immersed in different solutions as follow: A1 and A2 indistilled water (control); B1 and B2 in black tea; and C1 and C2 in tomato sauce for half an hour/day for seven days.Using a colorimeter, Teeth color measurements were recorded at baseline, after bleaching, staining, and polishing.Color changes were recorded according to the Vita shade guide and the CIE Lab system. Student's t-test was usedto analyze differences between the subgroups at p<0.05.Results: Significant color changes were recorded for the tea group after staining, but not after polishing (p<0.05). Nosignificant differences in color measurements were recorded between the subgroups of each group at all periods(p>0.05).Conclusion: Only tea produced clinically perceivable color change of bleached enamel after staining as well asafter polishing. MI Paste Plus did not affect enamel color change for all the groups.

Abstract

Aim of the study: Is to evaluate the effect of myrrh oil local application on the healing process of skin wounds histologically , histomorphometrically and , histochemically.Materials and methods:Twenty male white New Zealand rabbits were used in this study. An incisional wounds with full thickness depth and of 2 cm length were done on both sides of the cheek skin of each rabbit. The left sided incisions (the control group) were irrigated with distilled water (10µL). The right sided incisions (the experimental groups) were treated with myrrh oil (10µL). Each group was subdivided into 4 subgroups according to the healing interval into 1,3,7 and 14 days(5 rabbits for each group). Results: Histological findings of our current study showed a highly significant difference between the experimental and the control groups in count of the inflammatory cells which showed that mean values increased with time for the control and the experimental groups. The histomorphometrical findings had shown that the thickness of the epithelium was nearly completed at about 7 days for the experimental groups and at about 14 days for the control. The blood vessels count was recorded to have a highly significant difference between the groups at days 1 and 3 only. The histochemical findings had shown that the collagen fibers remodeling had recorded a highly significant difference between the control and the experimental groups at days 7 and 14.Conclusion: The current study had revealed that myrrh oil accelerates wounds healing in rabbits.

Abstract

AbstractTo investigate the characteristic intensity and pattern of staining of P53 and Ki67in oral squamous cell carcinoma in different grading of lesions.Thirty patients with oral squamous cell carcinoma were collected and used todetect the intensity and pattern of staining of P53 and ki67 markers byimmunohistochemical method.Immunoreactivity of this study for P53 found that 21 (70%) were positive indifferent grading of disease. Most frequent combination includes strong intensity in11 cases (52%) and diffuse pattern in 15 cases (71%).Immunoreactivity of Ki67 in oral squamous cell carcinoma indifferent gradingwas found positivity for ki67 in 25 cases (83%) and the most frequent pattern ofstaining was the scattered which seen in 20 cases (80%) and 5 cases (20%) had diffusepattern of staining.In conclusion the study revealed that the intensity and pattern of staining of P53and Ki67 correlate with well differentiated oral squamous cell carcinoma.
